چکیده

IN EARLIER work from this laboratory it was shown that arylsulphatase of chicken brain resembles arylsulphatase A of other animal species in several of its properties but exhibits certain characteristics similar to that of arylsulphatase B (FAROOQUI and BACHHAWAT, 1971). Recently the arylsulphatase A of chicken brain was purified and it was demonstrated that the purified enzyme could desulphate cerebroside-3-sulphate also (FARWQUI and BACHHAWAT, 1972). In the present report we have made a study of the kinetic properties of this unique arylsulphatase A purified from chicken brain using p-nitrocatechol sulphate and cerebroside-3-sulphate as substrates.

The regional distribution, age dependent variation and species differences of brain arylsulphatases.

The relative proportions of arylsulphatase A and B were determined by the method of BAUM, DODGSON and SPENCER (1959) in brains of various animal species and it was found that there was a considerable variation in the concentration of these two enzymes. Arylsulphatase A and B of various animal species including rat, man, monkey, sheep and chicken were partially separated using zinc acetate fract...

Purification of soluble arylsulphatases from ox brain.

1. Two soluble arylsulphatases (A and B) have been extracted from ox brain by a modified Albers autolysis method and purified by acetone and ammonium sulphate precipitation and dialysis. 2. A 1600-fold purification was achieved with arylsulphatase A and 320-fold purification with arylsulphatase B. 3. The specific activity of arylsulphatase A was 266000 4-nitrocatechol units/mg. of protein N, an...

Frequency of arylsulphatase A pseudodeficiency associated mutations in a healthy population.

Arylsulphatase A (ASA, EC 3.1.6.1) is a lysosomal enzyme that catalyses cerebroside sulphate degradation. ASA deficiency is associated with metachromatic leucodystrophy (MLD), a rare autosomal recessive disorder, which is characterised by the storage of cerebroside sulphate. Low ASA activities can be also observed in clinically healthy persons, a condition termed ASA pseudodeficiency. Two mutat...

Metabolism of sodium oestrone ( 35 S) sulphate in the guinea pig.

Intraperitoneal administration of sodium oestrone [(35)S]sulphate to male and female free-ranging guinea pigs is followed by excretion of most of the radioactivity mainly as inorganic [(35)S]sulphate in the urine within 72h. The remainder of the radioactivity in the urine was found in oestrone [(35)S]sulphate, two unidentified metabolites (A and B) and traces of oestradiol-17beta 3-[(35)S]sulph...

Regulation of arylsulphatase A and sulphogalactolipid turnover by cortisol in myelinogenic cultures of cells dissociated from embryonic mouse brain.

Myelinogenic cultures of cells dissociated from embryonic mouse brain were used to study the regulation of myelination-associated molecules by cortisol. Cortisol in physiological concentrations (0.03 microM) caused an increased accumulation of myelination-associated sulphogalactolipids. It also stimulated the myelin- and oligodendroglia-specific cyclic nucleotide phosphohydrolase. The increase ...
